Misra is a microbial geneticist who studies how a bacterium like Escherichia coli responses and endures various stresses, including those caused by antibiotics, defective envelop assembly, protein misfolding, heat, and osmotic-shock. Employing various genetic selection strategies, his lab has isolated several stress-alleviating mutations in a gene (rpoB) coding for an essential RNA polymerase beta-subunit. His current research involves investigating the mechanisms by which mutations in RNA polymerase overcome multiple and seemingly unrelated stresses.
A second project in his laboratory examines the mechanism of drug resistance, with an emphasis on the major multi-drug efflux pump system of clinical relevance. His research employs classical genetics and molecular biology approaches to investigate pathways of drug import and export that are crucial for drug resistance.
